Protocellular CRISPR/Cas‐based Diffusive Communication Using Transcriptional RNA Signaling

Protocells containing enzyme-driven biomolecular circuits that can process and exchange information offer a promising approach for mimicking cellular features developing molecular platforms. Here, we employ synthetic transcriptional together with CRISPR/Cas-based DNA processing inside semipermeable protein-polymer microcompartments. We first establish protocell be activated by external strands produce functional RNA aptamers. Subsequently, engineer module to generate functioning as diffusive signals sensed neighboring protocells trigger the activation of internalized probes or localization Cas nucleases. Our results highlight opportunities combine CRISPR/Cas machinery nanotechnology protocellular communication provide step towards development capable distributed processing.
